In forensic science, DNA plays a significant role, as it can directly aid in solving crimes and assisting the court. Even from highly decomposed bodies, DNA can be extracted and can be used to generate a complete profile. The study aims to examine the recovery rate and degradation rate of DNA from ligatures tied to the buried porcine cadavers. Therefore, the review is based on examining the recovery rate of DNA from human remains, as well as the decomposition stages and effects of decomposition on DNA. The review systematically provides the status of research on DNA recovery and analysis, and assess the factors influencing DNA preservation, extraction techniques, and forensic utility. Many platforms were used to search the database related to forensic science, molecular biology, and taphonomy. Studies were included that examined decomposition models, DNA recovery from human remains, and the impact of environmental factors on DNA deterioration in burial settings. The findings demonstrate that it is possible to extract DNA from highly decomposed bodies. However, the most significant finding is that no single study has discussed the extraction of DNA from ligatures found on buried or decomposing bodies. If proper collection and analytical techniques are employed, DNA analysis of ligatures from burial contexts can be a viable forensic option. The only concern is that applying optimal extraction procedures and comprehending material-environment interactions are essential for success.
